Abstract

Coffee consumption has been trending in our society due to numerous newly emerging coffee shops. Coffee shops nowadays serve various types of coffee to fit the taste of society so that they will consume according to their preferences. Coffee contains caffeine, chlorogenic acid, diterpenes, and other substances. The substances contained in coffee, when consumed, may have positive and negative effects on the body. However, there has been no research which discusses the safe limits of consumption of Robusta Coffee (Coffea anephora) and its impact on the kidneys. This study aimed to examine the effect of a single dose of oral acute toxicity test of Lampung Robusta coffee bean extract (Coffea canephora) on kidney histopathological structures of Sprague-Dawley rats (Rattus norvegicus) using the OECD test guideline No. 423. Coffee extract as a test material was given orally once to test animals with predetermined doses (fixed dose) based on the OECD guideline No. 423, which are 5, 50, 300, and 2000 mg/kgBW. Kidney histopathological observations according to the OECD guideline No. 423 were carried out in the control group, 300mg/kgBW, 2000mg/kgBW, and 5000mg/kgBW were assessed by a scoring system. The average results of histopathological observations of kidney cells in the control group, 300mg/kgBW, 2000mg/kgBW, and 5000mg/kgBW were 3.3%, 20.6%, 50.3%, and 72.3%. Analyzes were conducted with Kruskal Wallis and Post-Hoc Mann-Whitney, and significant results were obtained between the control group with 2000mg/kgBW and the control group with 5000mg/kgBW. There is an effect of oral acute toxicity test of Robusta coffee bean extract (Coffea canephora) Lampung on kidney histopathological description of male white rat (Rattus norvegicus) Sprague-Dawley strain based on OECD test guideline no. 423.
